The clear tube is the indicator of how much water (with or without additive, the blue by the way goes in the waste tank, pink in the flush tank) is left in the flush tank.
Unclip the tube and lower it to point down and it will empty the flush tank.

I'm assuming that the clear tube is the 'sight glass' for the flush water.
1. Have you actually put water in the flush tank, either from above through the toilet roll holder hole or from outside into the swivelling filler on the end of the cassette tank, again, assuming that this is a C200 based toilet?
2. Is the habitation 12V switched on, the flush usually won't work if not?
The flush should be operated by pushing down on the big button, turning it opens and closes the cassette flap to allow 'doings' to enter the cassette and to seal it again when finished.
TIP: Don't do any 'doings', even liquid ones, into the bowl with the flap closed. The liquid will run onto the top of the cassette making a nasty smelly mess if the lip seal on the flap is worn.
Ask me how I know this...

Offside 'wall' panel near the cooker/sink. It's a small panel approx 6 x 4" with four switches.....Master/Battery and Water/Pump/Lights. You need to turn the Master switch on.....ie, making sure it's switched to 'reset' position. All switches in caravans and campers work opposite to domestic household so the "off" position is actually "on".

Forgot to add....that cover which turns left/right opens your cassette flange when you use the toilet. Press down to flush. The clear plastic tube is simply an indicator when filling the toilet tank. It pulls out of the clip and to drain off you just pull it from the pipe end.
